Fava Bean (Vicia faba) Hydrolysate (as PeptiStrong™)

PeptiStrong™ is an ingredient derived from fava beans that contains extremely beneficial natural compounds called bioactive peptides. Other sources of naturally occurring bioactive peptides are collagen, insulin, and glutathione, all of which have crucially beneficial properties. These peptides have undergone patent protection and extensive research, including both pre-clinical and clinical studies. The studies have shown positive effects on gene expression and the secretion of tumor necrosis factor alpha (TNF-a). In vitro experiments using murine skeletal muscle cells and human macrophages have validated the ability of the peptides within PeptiStrong™ to increase muscle protein synthesis and reduce TNF-a secretion. The study also assessed the bioavailability and stability of the peptides, which proved to be resistant to digestion, displayed good intestinal barrier transport, and remained stable in plasma.

In an in vivo study using a murine model of muscle atrophy, PeptiStrong™ treatment for 18 days resulted in a significant attenuation of muscle loss and increased expression of muscle fibers. The treatment also upregulated protein-synthesis-related genes in muscle tissue. Other protein hydrolysates of fava bean and food-derived hydrolysates did not exhibit the same level of efficacy in vitro. Thus, PeptiStrong™ demonstrated efficacy in attenuating muscle atrophy through various mechanisms, including inflammation reduction and muscle protein synthesis.

A clinical study compared the effects of PeptiStrong™ and milk protein supplementation on muscle mass and strength loss during limb immobilization. The study involved 30 young men subjected to one-legged knee immobilization followed by recovery. PeptiStrong™ supplementation performed similarly to milk protein in modulating muscle size loss during immobilization and regain during remobilization. Notably, higher muscle protein synthesis rates were observed during remobilization in the PeptiStrong™ group.

Another study with 30 healthy male subjects investigated the effects of PeptiStrong™ supplementation on strength recovery and fatigue. The subjects were divided into groups receiving either a placebo or 2.4 grams of PeptiStrong™ daily. Delayed onset muscle soreness (DOMS) was induced using resistance exercise, and the subjects' strength recovery and fatigue were measured over three days. PeptiStrong™ supplementation significantly improved strength recovery, reduced fatigue, and positively influenced markers associated with muscle homeostasis. Myostatin expression was also found to be significantly reduced, indicating an anabolic response following intense exercise.

Maral Root Extract (Rhaponticum carthamoides) (10:1)

Maral Root Extract, derived from the plant Rhaponticum carthamoides, is a natural ingredient highly regarded for its adaptogenic properties and numerous health benefits. Maral Root Extract has been found to increase ATP production, which can allow individuals to exercise for longer periods and delay the onset of fatigue.

Furthermore, studies suggest that Maral Root Extract supplementation may promote greater muscle protein synthesis. While the exact mechanisms of 20-hydroxyecdysone (a plant ecdysteroid) on muscle protein synthesis are not fully elucidated, it appears to involve a rapid increase in calcium in the cell (cytosol) causing activation of Akt/PI3K signaling. This signaling promotes the metabolism, proliferation, cell survival, growth, and angiogenesis of cells. Along with these increases in muscle protein synthesis, Maral Root Extract has also been shown to aid in muscle recovery and improve muscular strength, enabling individuals to generate more power during workouts.

Moreover, the active compounds present in Maral Root Extract, such as ecdysteroids and additional flavonoids, exhibit potential anti-inflammatory and antioxidant effects through their adaptogenic properties. These proposed benefits listed above and the compelling research surrounding them has led epicatechin to be involved in innovations where muscle growth and improved performance are the goal.

How does epicatechin work? Glad you asked. The proposed mechanism of action of epicatechin is through the inhibition of myostatin, which is a type of protein called a myokine that in essence acts as a regulating molecule released by muscles. Increased levels of myostatin have been linked to limiting the level of muscle growth. To combat an increased concentration of myostatin, epicatechin can work to increase another protein called follistatin which the body can then use to control metabolism, promote muscle growth, and boost fertility. Increased myostatin has been linked to the clinically obese and inactive as well as individuals who have some degree of insulin resistance. Studies have shown that aerobic activities, strength training, and some modifications to diet can, to a degree, decrease myostatin levels in muscle. When it comes to epicatechins ability to increase follistatin levels and further inhibit myostatin concentrations as few studies have been conducted for its efficacy.

One study conducted on males showed that just 170mg of epicatechin per day was able to increase follistatin by about 50% and decrease myostatin by about 17%, which also coincided with an increase in strength of about 7%.

Looking at epicatechins nitric oxide boosting benefits, one animal study showed that nitric oxide and endurance was able to be increased even in the absence of exercise. This can aid in nutrient absorption and improved heart and muscle health outside of the gym but in the gym, it can also result in greater blood flow to muscles and stimulate a better pump in response to training.

The literature that has been presented on epicatechin in in vivo studies on muscle growth potential and improved overall health is promising and when combined with the other ingredients in this product, the body can be set up for the most beneficial environment for muscle growth, strength increases, and increases in performance.
